VisiATP - Engineered Nucleotides for Biomolecule Labeling
Take control of your labeling reaction!
With the new VisiATP biomolecule labeling product, you can efficiently label
DNA, RNA, or oligos with a fluorescent dye (or biotin) for use in various
applications with a simple one-step reaction. This flexible process allows you
to label what you want, when you want, with the fluorescent dye of your choice.
An established process, adapted for fluorescence.
VisiGen has taken advantage of the consistent, reliable “forward reaction” that
is commonly used to enzymatically transfer a 32-P gamma-phosphate of an ATP
molecule to the 5’-OH of a nucleic acid molecule, and adapted it for use with
fluorescence. The keys to this simple, one-step system are novel ATP
nucleotides engineered by VisiGen with an attached fluorophore or biotin moiety
at the gamma-phosphate position. These engineered nucleotides allow the direct
addition of a fluorescent dye (or biotin) to the 5’ end of a DNA or RNA
strand.
5’ end labeling with fluorescence makes sense.
Fluorescence-based technologies have emerged as the methods of choice over
radioactivity for monitoring biomolecules. Increasingly sensitive assays have
enabled a switch from potentially harmful radioactivity to the more
environmentally friendly fluorescence. Labeling nucleic acids at either the 5’
or 3’ end has the advantage that a single fluorophore is incorporated per
molecule making quantitation in downstream assays straightforward. In addition
to problems with sequence bias, labeling nucleic acids with multiple internal
modifications can affect their hybridization efficiency and make quantitation
difficult. 5’ end labeling naturally leaves the 3’ end free for subsequent
enzymatic reactions.
Other Applications.
VisiGen's novel phosphate transfer technology also has
applications for protein labeling, ATP binding studies, and phosphate transfer
reaction monitoring.
